That was one detail not stated.
Suppose your brother decides that he, in turn can lend the car to yet another party - and they either wreck it, or engage in behavior that gets it seized?
The agreement was for you and your sister to alternate. It didn't provide for a third party. If she is loaning it out to someone else, she is in violation of the agreement. This is like a partnership, where one party can get the other into financial (and even legal) difficulty without their knowledge of consent.
Either re-state the agreement in writing this time, or dissolve the agreement and one buys out the other, or sell it and split the proceeds.

The biggest issue is the liability. You two can drive the car, but no one else should. It would make for a legal nightmare if someone else, who is not a legal owner, gets in an accident. You could be held responsible too even if it was your sister's "weekend". If she doesn't need the car anymore, buy her out, end the headache and preserve your relationship with your sister.

Do you really want to spend your life micro managing your sister? Trust me, that will be completely exhausting and will have little effect. Your sister's weekends are her weekends and you are not going to be there to see who is driving and who is not. Make sure she knows that her weekends are her responsibilty and even sign something legal to the effect that she is financially responsible to fix the car if you are worried about it, but then trust her and that she will make wise decisions because it is her transprotation too. Remember though: accidents do happen and they could happen to you too. Buckle up.
